摘要
目的:采用戊型肝炎病毒主要免疫表位多肽检测特异性IgM抗体。方法:采用戊型肝炎病毒主要免疫表位多肽,制备ELISA检测试剂,同时使用北京万泰生物药业公司生产的抗-HEV IgM抗体诊断试剂,分别检测33例戊型肝炎患者血清和344例普通人群血清抗-HEV IgM,并对部分阳性标本用免疫印迹法(Western blot,WB)确认。结果:用本室ELISA试剂检测33例戊型肝炎患者血清,抗-HEV IgM均阳性,符合率为100%;用万泰公司试剂检测阳性31例,符合率为93.9%。344例普通人血清中,本室试剂检测抗-HEV IgM阳性11例,而用万泰试剂检测仅其中6例显示阳性;对本室试剂阳性而万泰试剂阴性的5例进一步用WB检测,抗-HEV IgM均阳性。结论:本室制备的ELISA试剂可准确高效地检测血清抗-HEV IgM。
Objective:To detect specific IgM antibodies against hepatitis E virus in human serum by ELISA based on the immunodominant polypeptide of ORF2.Methods:Anti-HEV IgM in serum samples from 33 acute hepatitis E patients and 344 general persons was detected in parallel by ELISA developed in this study and by the kit prepared by Beijing Wantai Biological Pharmacy Enterprise Co.,LTD.Western blotting was used to confirm the antibody status.Results:All the 33 serum samples from acute hepatitis E patients were positive for anti-HEV IgM detected by our ELISA and the accordant rate was 100%.Thirty-one of them were positively detected by Wantai ELISA kit and the accordant rate was 93.9%.In 344 serum samples from general population,our ELISA showed that 11 were positive.However,Wantai kit showed that only 6 of them were positive.Western blot analysis clarified that the remained 5 samples were all positive for anti-HEV IgM.Conclusion:The ELISA based on the immunodominant epitope of ORF2 developed in this study is highly sensitive and specific for detection of anti-HEV IgM.
