安律胶囊对豚鼠心室肌细胞钾离子通道的影响 被引量:3

摘要 目的:通过研究安律胶囊对豚鼠心室肌细胞钾离子通道的影响,以探讨其抗心律失常的作用机制。方法:采用膜片钳全细胞记录技术记录了安律胶囊对单个豚鼠心室肌细胞钾电流的影响。结果:①安律胶囊清膏2、20、200μg/L可使IK1幅值从给药前的(-2.06±0.29,nA)降低到(-1.95±0.34、-1.67±0.43和-1.62±0.25,nA),大剂量具有显著的统计学意义(P<0.05);在对IK1时效关系的实验中,在200μg/L的安律胶囊清膏作用下,未计时时最大内向峰值钾电流为(-1.60±0.27,nA),1min、4min、8min后,最大内向峰值钾电流分别为(-1.56±0.35,-1.34±0.26和-1.26±0.29,nA),无统计学意义(P>0.05)。②安律胶囊清膏2、20、200μg/L可使Ito1幅值从未给药时的(1.14±0.17,nA),降低到(1.13±0.17、1.10±0.16和1.06±0.13,nA),但无统计学意义(P>0.05)。③安律胶囊清膏2、20、200μg/L可剂量依赖性的降低Ik,分别使Ik幅值从给药前的(1.45±0.15,nA),降低到(1.42±0.15、1.29±0.10和1.20±0.11,nA),中剂量组具有显著的统计学意义(P<0.05),大剂量具有极为显著的统计学意义(P<0.01);在对Ik时效关系的实验中,在200μg/L的安律胶囊清膏作用下,未计时时最大内向峰值电流为(1.45±0.15,nA),1min、4min、8min后,最大内向峰值钾电流分别为(1.21±0.10,1.20±0.12和1.11±0.13,nA),无统计学意义(P>0.05)。结论:安律胶囊对IK1有一定的阻滞作用且可剂量依赖性的阻滞Ik,但不具有时间依赖性,对Ito1无阻滞作用,这是其抗心律失常作用的机理之一。 Objective: To study the ANLV capsule on guinea pig ventricular myocytes the effects of potassium ion channels in order to investigate the role of anti-arrhythmic mechanism. Methods :The wholecell patch-clamp recording technology records law capsule on an individual guinea pig ventricular myocytes the effects of potassium currents. (2) Clearance paste of ANLV capsules 2,20,200 μ g / L can make hol administration at the time of the amplitude (1.14 ±0.17, nA), decreased to (1.13 ±0.17,1.10±0.16 and 1.06 ±0.13, nA), but no significant (P〉0.05).(3) Clearance paste of ANLV capsules 2,20,200 μg / L may be dose-dependent reduction of Ik, respectively Ik amplitude from pre-administration (1.45 ± 0.15, nA), decreased to (1.42 ± 0.15,1.29 ±0.10 and 1.20 ± 0.11,nA), middle dose group with significant statistical significance (P〈0.05), large doses of great significant statistical significance (P〈0.01); statute of limitations on the relationship between Ik experiments, in 200 Ix g / L of On the role of law under clearance paste of ANLV capsules, no maximum time to peak current (1.45 ±0.15, nA), 1 min, 4min, 8min, the largest peak inward potassium current, respectively (1.21 ± 0.10,1.20 ±0.12 and 1.11± 0.13, nA), no statistical significance (P〉0.05).Conclusion: ANLV capsule have a role of a certain block forIK1
