摘要
目的探讨在家族性乳腺癌中影响易感基因(BRCA1)蛋白表达的因素。方法运用免疫组织化学SP法检测10例乳腺导管内乳头状瘤和8例遗传性乳腺癌组织中BRCA1的表达。结果在10例乳腺导管内乳头状瘤病例中,BRCA1蛋白阳性位于细胞核,阳性表达率为100%(10/10);8例遗传性乳腺癌组织中BRCA1蛋白阳性表达位于细胞浆或细胞核和细胞浆,阳性率为50.0%(4/8),两者差异有统计学意义(P<0.01)。可能由于标本数量较少,BRCA1在伴有腋窝淋巴结转移组的失表达率与无淋巴结转移组差别无统计学意义(P>0.05)。结论在家族性乳腺癌中,BRCA1的失表达明显高于乳腺良性肿瘤。
Objective To investigate the expression of BRCA1 protein in inherited breast cancer. Methods The expression level of BRCA1 protein was detected by immunohistochemistry (IHC) with Streptavidin-Peroxidase (SP) method in benign breast tissues and inherited breast carcinoma tissues. Results The positive expression rates of BRCA1 were 100 % (10/10) and 50.0 % in benign breast tissues and inherited breast carcinoma tissues, respectively. The difference in BRCA1 expression between the two groups was significant (P〈O. 01). In benign breast tissues, BRCA1 express in nuclei while it express in cytoplasm or in cytoplasm and nuclei in the breast cancer tissues. Loss of BRCA1 expression in breast cancer tissues was not significantly related to metastasis to axillary lymph nodes. Conclusion Loss of BRCA1 expression in inherited breast cancer is higher than that in benign breast tissues.
