摘要
目的研究高效液相色谱快速测定沙地濒危植物长柄扁桃仁中α-维生素E(α-生育酚)含量的方法。方法采用色谱柱为Shim-pack VP-ODS柱(150mm×4.6mm,5iμm);流动相为甲醇-水(体积比为98:2),流速1.2mL/min;检测波长为300nm;柱温为室温。以保留时间定性。峰面积定量,标准曲线法计算样品中α-维生素E(α-VE)的含量。结果方法的线性范围为1.298~103.84μg/mL,线性相关系数为0.9999;检出限为0.19μg/mL;精密度为1.21%~3.26%;方法回收率为84.93%~90.53%,平均回收率为88,15%(RSD=2.68%)。测得长柄扁桃仁中α-VE含量为2.82μg/g,α-VE,δ-VE,β,γ-VE的相对百分含量分别为0.94%,92.78%,6.28%。结论长柄扁桃仁具有一定的经济价值,从中提取的天然维生素E为低α型混合生育酚,在医药、食品、饲料、化妆品方面可作为一种抗氧化剂.为全面评价长栖扁桃仁的营幕品盾及为产品开发利用提供科举依据.
Aim To provide scientific supports for comprehensive assessment and utilization of the nutlet oF endangered plant long carpopodium almond (Amygdalus pedunculatus Pall. ) by developing a HPLC method for the determination of alpha tocopherol content in this nutlet. Methods Alpha tocopherol was separated by employing a column of Shim-pack VP-ODS ( 150 mm ×4. 6 mmi. d. ,5 μm) and a mobile phase of methanol-water(98:2 ,v/v) at a flow rate of 1.2 mL/min and room temperature with were used for the qualitation and quantitation of alpha UV detection at 300 nm. Retention times and peak areas tocopherol, respectively. Results Linearity was confirmed in the mass concentration range of 1. 298 ~103.84 μg/mL with the correlation coefficient of 0. 9999 and the limit of detection(LOD) was 0. 19 μg/mL. The relative standard deviation (RSD) was in the range of 1.21% ~3.26%. The recoveries obtained ranged from 84.93% to 90. 53% and the average recovery was 88.15%. The alpha tocopherol content of long carpopodium almond nutlet was 2. 82 p,g/g and the relative percentage content of tocopherol isomers were 0. 94% , 92. 78%, 6.28% , respectively. Conclusion The natural tocopherol extracted from long carpopodium almond nutlet was mixed tocopherol, it had low content of alpha tocopherol and high content of tocopherol isomers. Therefore it has a certain economic value and could be used as a kind of antioxidant in the following fields, such as medicine, food, feedingstuff and cosmetic.
