摘要
以6-甲基-2,4-二羟基嘧啶(6-MU)在无水乙醇中与甲醛和二乙胺反应,合成了5-二乙氨甲基-6-甲基-2,4-二羟基嘧啶。考察了影响反应收率的各种因素,确定了优化的反应条件,即:n(6-MU):n(二乙胺):n(CH2O)=1.0:2.0:2.0,反应溶剂无水乙醇,6-MU的投料量为0.02mol的情况下,溶剂量60mL,反应时间3h,产物收率34.9%。质谱和1HNMR对产品进行了结构鉴定,确定所得产品为目标物。
6-methylpyrimidine-2,4(1H,3H)-dione was reacted with diethylamine and formaldehyde in anhydrous ethanol, and 5-diethyl aminomethyl-6-methyl-2,4-dihydroxypyrimidine was generated. Various factors affecting the reaction yield were studied and the optimum reaction conditions was determined as follows: 6-methylpyrimidine-2,4(1H,3H)-dione: diethylamine: for-maldehyde(molar ratio)= 1.0.:2.0:2.0, with anhydrous ethanol as solvent, the charged amount of 6-MU was 0.02 mol, and the solvent volume is 60 mL, reaction time is 3 hours, and the yield of the final product is 34.9%. Structural identification was carried out through LS-MS and 1HNMR detection and the product was confirmed as the goal product 5-diethylaminomethyl-6-methyl-2,4-dihydroxy pyrimidine.
