黄柏、苍术药对提取物中盐酸小檗碱在大鼠体内的药代动力学研究 被引量:12

摘要 目的:黄柏、苍术药材按1:7配比混合,经提取浓缩得到的提取物,研究其中所含的盐酸小檗碱在大鼠体内的药代谢动力学参数,以揭示药对提取物中盐酸小檗碱在大鼠体内的变化规律。方法:以吴茱萸碱为内标物,利用HPLC测定血液中盐酸小檗碱的含量。结果:大鼠血浆样品中,盐酸小檗碱的线性范围为0.006 6~0.211 2μg/mL,提取回收率较高,日内日间精密度RSD均小于10%,检测限为0.003 3μg/mL,主要药动学参数如下:Tmax(h)为2.000,Cmax(mg/L)为0.183,MRT0-24(h)为10.867,MRT0-∞(h)为21.562±4.448。结论:药对提取物中盐酸小檗碱在体内吸收较快,但代谢较慢。 AIM: To use seven parts cortex phellodendri and one part Rhizoma atractrylodis to extract mixedly,and study mouse s pharmacokinetic parameter of berberine derived from extract. METHODS: Evodiamine was taken as the marker,HPLC was used to determine berberine content in rat plasma. RESULTS: The linear range of rat s plasma sample was within 0.006 6-0.211 2 μg/mL,RSD was little than 10%,detection limit was 0.0033 μg/mL.The main pharmacokinetic parameters consisted of 2 h(Tmax),0.183 mg/L(Cmax),10.87 h(MRT.24),21. 562 ±4. 448 (MRT0-∞). CONCLUSION: Berberine in combination of cortex phellodendri and Rhizoma atrac- tylodis can be quickly absorb and achieve the peak value, but metabolize slowly.
