摘要
用人工流产胎皮提取人Ⅲ型前胶原（hPcⅢ）免疫BALB／C小鼠，成功地得到抗hPcⅢ单抗2株即MA1和MH8。杂交瘤连续培养半年，冻存5年复苏后仍能分泌抗体。经免疫级化检测显示成人血管壁、纤维间质和中国仓鼠成纤维细胞株（V79）呈阳性反应、肝癌细胞及问质也呈阳性反应，其余呈阴性。MA与牛Ⅰ型、Ⅲ型胶原及角蛋白有交叉免疫反应。MH8与牛Ⅰ型、Ⅲ型胶原有交叉，与角蛋白无交叉。两株单抗均与甲胎蛋白（AFP）、癌胚肮原（CEA）无交叉免疫反应。MA1属IgM型，其杂交瘤染色体数96条，MH8属IgG1型，其杂交瘤染色体效90条。
Human procollagen type Ⅲ (hPCⅢ )was isolated and purified from fetal human skin. BALB/C mice wereimmunized witb hPCⅢ. Hybridoma which secretes monoclonal antibodies against hPCⅢ have been established and twoof the monoclonal antibodies have been detected by enzyme-linked immunosorbent assay (ELISA)and immunohistology.The result suggested that the monoclonal antibodies might be suitable to immunohistochemical studies as well as to im-munoassays for PC Ⅲ.
