摘要
目的建立一种大蒜总皂苷含量的测定方法。方法以薯蓣皂苷元为对照品,香草醛-冰醋酸-高氯酸比色法测定大蒜总皂苷含量。结果对照品在1.67-13.34μg/ml范围内与吸光度呈良好线性关系,回归方程为:A=0.014C-0.012(R=0.999),平均回收率97.8%,相对标准偏差(relative standard deviation,RSD)=1.98%。方法学考察各项指标均符合要求。结论总皂苷测定方法简单,准确性、稳定性和重复性良好,可作为大蒜总皂苷质量控制手段。
Objective To develop a method for content determination of total saponins in garlic.Methods Diosgenin was used as a reference substance,and vanillin-perchloric acid was applied to determine the content of saponins at 532 nm.Results The calibration curve of diosgenin showed good linearity in the range of 1.67~13.34 μg/ml,and the regression equation was A=0.014C-0.012,R=0.999.The average recovery rate of control compound was 97.8%,and relative standard deviation(RSD)was 1.98%.All the indexes of the methodological investigation met the requirements. Conclusion The determination method is simple, rapid, sensitive and accurate, which can be used for evaluating the extraction quality of total saponins.
