摘要
目的研究曲霉3277次级代谢产物中抗结核活性成分。方法通过DIAION HP20大孔树脂吸附、Toyoperal HW-40F凝胶以及硅胶柱层析,结合生物自显影,对曲霉3277发酵液中的抗结核分枝杆菌活性成分进行分离,采用NMR、ESI-MS技术鉴定其结构,并采用MABA方法,对结核分枝杆菌H37Rv最低抑制浓度(MIC)进行测定。结果分离得到了3个化合物,分别命名为3277-1、3277-2、3277-3。其中的一个化合物3277-2的结构为:4-((1Z,3Z)-2,3-二异腈基-4-(4-甲氧基苯基)1,3-丁二烯)硫酸苯酯钠。3277-2对结核分枝杆菌H37Rv的MIC为8μg/ml。结论3277-2为新化合物,体外具有明显的抗结核活性。
Objective To study the second metabolites with anti-tuberculosis activity produced by Aspergillus sp. 3277. Methods Bioautography-guided isolation with macroporous adsorption resin diaion HP20, toyoperal HW-40F gel and silica gel column chromatography were used to separate and purify the compound. NMR and ESI-MS were used to identify its structure. Results Three compounds named 3277-1,3277-2 and 3277-3 were separated from the fermentation broth produced by epiphyte 3277. 3277-2, one of the three compounds, was concluded as sodium 4- ( ( 1 Z, 3Z)-2,3- diisocyano-4-(4-methoxyphenyl)buta-1,3-dienyl)- phenyl sulfate. The MIC on tubercle bacillus H37 Rv was 8μg/ml . Conclusion 3277-2 was a new compound with notable anti-tuberculosis activity.
