姜黄素对肺纤维化鼠Bcl-xl表达的影响

The Expression of Bcl-xl and Apoptosis in the Epithelial Cells of Pulmonary Fibrosis Rats and Intervention Study of Cucumin

摘要 目的探讨姜黄素对博莱霉素诱导肺纤维化大鼠肺组织Bcl-xl基因表达的影响。方法选取SD大鼠96只,随机分为空白组、模型组、醋酸泼尼松龙组、姜黄素组,每组各24只。各组动物于7、14、28 d随机处死8只,取肺组织采用RT-PCR检测Bcl-xl mRNA的表达和免疫组化法检测蛋白的表达,观察姜黄素对博莱霉素诱导肺纤维化大鼠肺组织Bcl-xl基因表达的影响。结果模型对照组Bcl-xl蛋白与Bcl-xl mRNA表达在第7天开始下降,第14天达最低峰,第28天略有回升,但仍低于空白对照组,姜黄素组Bcl-xl mRNA及蛋白表达显著高于模型对照组及空白对照组,均有统计学意义(P<0.01);姜黄素组与泼尼松龙组比较7、14 d均无统计学意义(P>0.05),28 d姜黄素组Bcl-xl表达高于泼尼松龙组,有统计学意义(P<0.01)。结论在博来霉素诱导肺纤维化形成阶段应用,姜黄素干预可能通过上调Bcl-xl mRNA及蛋白的表达减轻肺纤维化的程度。 Objective To examine the expression of Bcl - xl in epithelial cells of pulmonary fibrosis rats with bleomycin -treated on different times and compared with the saline control group and with the group treated with the cucumin , in order to explore the effect of cucumin on Bcl - xl expression in the lung of rats with experimental pulmonary fibrosis. Methods 96 sprague dawley (SD) were randomly divided into four groups, normal control group, model group , control group, group treated with the cucumin, the group treated with acetic acid Prednisone. On the 7th, 14th, and 28th day the lungs were isolated. Pathological changesin the lungs were evaluated by HE stain. The mRNA expression of Bcl - xl was evaluated by RT - PCR quantitative analysis, and the immunohistochemical method was used to investigate the change of and Bcl - xl protein. Results The result of immune histochemistry and RT - PCR suggested that the expression of Bcl - xl protein and mRNA also decreased after 7 d by treated with bleomycin ,reaches the trough in the 14th days. the expression of Bcl-xl in the treatment groups with cucumin were higher than the model group and the control group( P 〈0.01 ). It had no statistics significance in the 7 d, 14 d between the treatment groups with cucumin and with acetic acid Prednisone(P 〉 0.05 ). But There was significant difference of statistics between the treatment groups with cucumin and with acetic acid Prednisone in 28 d about the the expression of Bcl - xl(P 〈 0. 01 ). Conclusion Bcl - xl gene and protein down - regulated in lung tissue of pulmonary fibrosis, which may play an important role in the development of pulmonary fibrosis. Cucumin can reduce the expression of pulmonary fibrosis inducede by the bleomycin .
