摘要
报道了一种抗真菌治疗新药酮康唑的吸附行为及其测定。该药物在玻碳电极上预富集后,用示差脉冲伏安法(DPV)测定,其响应电流与药物浓度在2.0 × 10-10~ 1.0 × 10-5mol/L范围内呈线性关系;检出限为4.0 × 10-11 mol/L。可用于尿和血清样品中药物含量的测定。采用计时电量法(CC)研究了酮康唑在玻碳电极上的吸附行为,求出了吸附自由能,确定了吸附模式。采用整体电解法测定了电极反应电子数,并结合氧化前后紫外光谱的变化推导了反应机理。
A sensitive stripping voltammetry to determine the antifugal drug-ketoconazole was reported. Ketoconazole was preconcentrated on the glassy carbon electrode in an open circuit, stripped by differential pulse voltammetry in a blank solution. There was a linear relationship between the stripping current and the concentration of ketoconazole in the range of 2× 10-10-1× 10-5 mol/L. The detection limit calculated by three times signal to noise was 4 × 10-11 mol/L. The calibration curves of ketoconazole in urine and serum samples were established by this method. The adsorptive behavior of ketoconazole at the glassy carbon electrode was investigated by chronocoulometry. The adsorption corresponded to the Frumkin type isotherm, and the free energy of adsorption was calculated to be 3. 59 x 104 J/mol at 25℃. Bulk electrolysis method was used to measure the number of transferred electrons in the oxidative reaction, and the mechanism of electrode was suggested.
