摘要 通过构建肌肉组织cDNA文库分离到鳜肌球蛋白轻链2基因(MLC2),基因登录号为FJ428249.MLC2基因cDNA序列全长1206bp,编码区长度为579bp.MLC2基因开放阅读框编码170个氨基酸,具有EF-手相家族蛋白全部4个EF-手相结构.与已报道的其他动物MLC2相比较,所推导氨基酸序列同源性在66.3%~97.6%,其中与Ca2+结合区域非常保守,7种鱼氨基酸序列同源性为100%.采用实时荧光定量PCR方法对鳜MLC2发育表达分析表明,MLC2在原肠期开始有低量表达,与原肠期相比,尾芽期、肌肉效应期和仔鱼阶段MLC2表达量随发育阶段渐进而升高. The myosin light chain 2(MLC2) gene was isolated from the muscle tissue cDNA library of Siniperca chuasti. The length of the MLC2 cDNA is 1 206 bp, and the open reading frame is 579 bp. The MLC2 encodes 170 amino acids, which includes four EF-hand structures. Compared with the MLC2 of other animals reported, the homology of the deduced amino acids sequences was between 66.3% -97.6%. The primary structures of the Ca^2+ - binding domain were well conserved among the MLC2s of seven other fish species. The ontogenetic expression analysis by real-time PCR showed that the MLC2 mRNA was first detected in gastrula stage and its expression gradually increased from the tail bud stage to larval stage.
