盐酸哌甲酯对注意缺陷多动障碍大鼠Homer 1a表达的影响 被引量:1

摘要 【目的】研究Homer 1a与注意缺陷多动障碍的关系,为临床治疗提供理论依据。【方法】将20只注意缺陷多动障碍动物模型-幼年自发性高血压大鼠(spontaneously hypertensive rat,SHR)随机分为两组,分别给予盐酸哌甲酯(2 mg/kg)或生理盐水,分别利用RT-PCR和Western Blotting技术检测两组大鼠Homer 1a mRNA和蛋白表达差异情况。【结果】盐酸哌甲酯给药组前额叶、海马中Homer 1a mRNA和蛋白的表达较生理盐水对照组显著增加(mRNAt前额叶=2.41,t海马=2.68,蛋白t前额叶=3.21,t海马=4.07,P值均<0.05)。【结论】盐酸哌甲酯单次用药可以引起SHR大鼠前额叶和海马Homer 1a mRNA和蛋白表达的增加,提示Homer 1a可能参与了ADHD的病因及发病机制。 [Objective] To investigate the effect of Homer la on attention deficit/hyperactivity disorder(ADHD). [Methods] 20 animal models of ADHD-spontaneous hypertensive rats (SHR) were divided into two groups (10 rats in each), and given saline (SAL) or methylphenidate (MPH), respectively, to examine the effects of MPH on the expression of Homer la using RT-PCR and western blotting, respectively. [Result] By RT-PCR and western blotting detection, it was found that the mRNA and protein expressive levels of Homer la were increased by MPH compare to SAL (mRNA t=2.41,and 2.68,protein t=3.21,and 4.07,All P〈0. 05). [Conclusion] It is plausible that Homer la may be involved in the etiology and pathogenesis of ADHD.
