摘要
在大肠杆菌中表达的重组人ＴＮＦα衍生物（ｒｈＴＮＦαＤ１１ａ）以可溶性形式存在，诱导后的菌体超声破碎后，依次经阶段盐析、离子交换层析及凝胶过滤，最终获得的ｒｈＴＮＦαＤ１１ａ纯度达９９％。
The recombinant human TNF derivative rhTNFα D11a was expressed in E.coli as soluble form.Heat induced bacteria were supersonical under gone disintegration, saltingout, ionexchange chromatography and gel filtration successively. The purity of TNF derivative could reach 99%.Toxicological analysis on L929 cell line showed that the activity ratio is about 1×108U/mg protien.
