摘要 采用改进的碳二亚胺法,将半抗原氧氟沙星(OFL)与载体牛血清白蛋白(BSA)和卵清白蛋白(OVA)偶联,制备得到氧氟沙星免疫原OFL-BSA和包被原OFL-OVA。通过其紫外扫描光谱、SDS-PAGE凝胶电泳实验,表明半抗原和载体成功偶联。用OFL-BSA免疫BALB/C小鼠,间接ELISA测定多抗上清(pAb)效价,阻断ELISA鉴定其敏感性,交叉反应实验鉴定其特异性。结果表明,获得了高价、敏感、特异的pAb,这对抗氧氟沙星单克隆抗体(mAb)的制备具有重要意义。 Ofloxacin(OFL) was coupled with carrier protein bovine serum albumin(BSA) and ovalbumin(OVA) to prepare immunizing antigen and OFL-BSA and coating OFL-OVA by modified method of carbodiimide. UV scanning spectrum and SDS-PAGE indicated that the artificial antigens both OFL-BSA and OFL-OVA were successfully coupled. BALB/C mice were immunized with OFL-BSA, the titre of polyclonal antibody(pAb) was detected by indirect ELISA, the sensitivity of pAb was identified by blocking ELISA and the specificity of pAb was identified by the method of cross-reaction test. The results showed that sensitive and specific anti-OFL pAb have been produced in sera of immunized BALB/C mice, which made it significant to prepare monoclonal antibody of Ofloxacin.
