摘要
本文以沙地云杉成熟合子胚为材料,在LP培养基上附加2,4-D、6-BA和KT进行愈伤组织诱导和增殖培养,在LS培养基上附加ABA和PEG进行分化培养。测定了沙地云杉体细胞胚胎发生过程中不同阶段,各类型愈伤组织蔗糖酶活力的变化。结果表明:在诱导和增殖阶段,蔗糖酶活力的变化趋势相同。在第0d^6d蔗糖酶活力逐渐增加,到第6d时达到最高峰,第7d^12d蔗糖酶活力下降,12d后又有所回升;分化培养基上的愈伤组织蔗糖酶活力变化稳定。
In this paper, mature zygotic embryos ofpicea mongolica for the material, medium was LP with 2,4 - D ,6 - BA and KT for callus induction and proliferation, and chose LS with ABA and PEG for differentiation. Determined callus sucrase enzyme activity in its various stages of the differences callus types. The results show that in the phase of induction and proliferation the sucrase enzyme trended the same vitality. In section 0d - 6d sucrose enzyme activity gradually increased, to the first 6d are peaked. But in section 7d - 12d sucrose enzyme activity decreased, 12d and then rebounded; Sucrase enzyme activity of the callus on the differentiation was in stability.
