摘要
目的研究补肾中药对体外蛋白非酶糖化产物的抑制作用。方法选用终浓度为0.1 mmol/L的怀牛膝、淫羊藿、熟地黄、肉苁蓉4味补肾中药及氨基胍(AG)分别加入终浓度为200 mmol/L的葡萄糖、50g/L的牛血清白蛋白(BSA)糖化系统中,37℃温孵12周后,用荧光光度计测定晚期糖化终末产物(AGEs)的荧光度值(F),计算抑制率(IR)和相对IR。结果怀牛膝、淫羊藿、熟地黄对AGEs的IR、相对IR与AG比较差异无统计学意义(P>0.05),怀牛膝、淫羊藿、熟地黄与AG的抑制作用相同;肉苁蓉对AGEs的IR、相对IR与AG比较差异有统计学意义(P<0.05),肉苁蓉对非酶糖化抑制作用低于AG。结论补肾中药怀牛膝、淫羊藿、熟地黄对体外蛋白非酶糖化产物有直接抑制作用。
Objective To observe the inhibition effects of herbs invigorating the kidney on nonenzymatie glycosylation products of protein in vitro. Method Final concentration 0.1 mmol/L of Radix Aehyranthis Bidentatae, Herba Epimedii, Radix Rehmanniae preparata and Herba Cistanehes (four herbs invigorating the kidney) and AG were added to Diastatic system containing final concentration 200mmol/L glucose, 50g/L BSA. After 12 weeks of ineubation in 37, fluorescence value of AGEs was measured. At that time inhibition rate (IR) and relative IR were calculated. Result IR and relative IR of Radix Aehyranthis Bidentatae, Herba Epimedii and Radix Rehmanniae preparata for AGEs were not decreased in comparison with those of AG ( P 〉 0. 05 ). The inhibition of Radix Achy- ranthis Bidentatae, Herba Epimedii and Radix Rehmanniae preparata was same as that of AG. The inhibition of Herba Cistanches on nonenzymatie glycosylation products was lower than that of AG ( P 〈 0. 05 ). Conclusions Radix Achyranthis Bidentatae, Herba Epimedii and Radix Rehmanniae preparata with invigorating the kidney has direct inhibition on nonenzymatic glyeosylation products of protein in vitro.
