摘要
目的 探讨中国南方汉族人A及AB血型的基因亚型分布,为实现跨ABO血型器官移植提供理论依据。方法取健康南方汉族成人体检者A型血330份,AB型血140份,提取DNA,采用序列特异引物聚合酶链反应(PCR—SSP)检测其基因亚型。将4种引物混合物同步扩增,根据PCR—SSP反应产生的特异性阳性条带判定相应的基因亚型。结果A型样本分型成功327份,AB型样本分型成功139份。A型血基因亚型以A/A201、A/O及A201/O较常见,占95%以上,而A201/A201及A/A不到5%;AB型血中A/B及A201/B基因亚型所占的比例较为接近,分别为54%和46%。结论中国南方汉族人A型血存在多种基因亚型,其特点明显不同于欧美白人,不同亚型之间的抗原性差异尚需要进一步研究。
Objective TO investigate the genotypes of the ABO blood group in the Han nationality of Southern China. Methods Four hundred and seventy blood samples from unrelated Hart individuals of Southern China including 330 cases of group A and 140 cases of blood group AB, were genotyped by the polymerase chain reaction with sequence-specific primers (PCR-SSP). Genomics DNA were extracted and amplified by four sequence specific primers, and the typing results were identified corresponding to the special positive strip of PCR reaction. Results Three hundred and twenty seven group A samples and 139 group AB samples were genotyped successfully. In group A, genotypes of A/A201, A/O, A201/O accounted for 95% of total, while A201/A201 and A/A accounted for less than 5%. In group AB, the frequencies of gene subtype A/B and A201/B were 54% and 46% respectively. Conclusion The results show that the distribution of genotype frequencies of Han nationality is significantly different with that of Caucasians populations.
