积雪草酸对大鼠局灶性脑缺血再灌注损伤的保护作用研究 被引量:5

摘要 目的研究积雪草酸对大鼠局灶性脑缺血再灌注损伤的保护作用及其可能机制。方法Wistar♂大鼠随机分为6组:积雪草酸低、中、高剂量组(50、100、200mg·kg-·1d-1),阳性对照杏灵颗粒组(300mg·kg-·1d-1),模型组和假手术组;大鼠灌胃给药,1次/d,连续7d,末次给药60min后采用线栓法制作大鼠局灶性脑缺血再灌注模型,缺血2h再灌注24h后对大鼠神经功能进行评分,并测定脑梗死体积、脑含水量、脑组织中白细胞介素-β(IL-β)和肿瘤坏死因子-α(TNF-α)的含量。结果与模型组相比,积雪草酸能够显著降低脑梗死体积和脑含水量,减少脑组织中的IL-1β和TNF-α含量(P<0.01,P<0.05)。结论积雪草酸对大鼠局灶性脑缺血再灌注损伤具有显著的保护作用,其机制可能与降低脑组织中的IL-1β和TNF-α含量有关。 Aim To investigate the protection by asiatic acid against focal cerebral ischemia-reperfusion injury in rats and the possible mechanism.Methods Male Wistar rats were randomly divided into six groups:low,middle and high dose of asiatic acid groups(50,100,200mg·kg-1·d-1),positive control group administered with Xingling Granule(300mg·kg-1·d-1),model group and sham operated group given with distilled water.All rats were intragastrically administered once a day lasted for 7days.Focal cerebral ischemia was induced at 1h after the last dose by middle cerebral artery occlusion(MCAO)using the method of intraluminal thread.Twenty-four hours after reperfusion foclouing two hours of ischemia,the neurological deficit scores,brain water content,infarct volume and changes in the concentration of interleukin-β(IL-β)and tumor necrosis factor-α(TNF-α)in brain tissue were determined.Results Compared with the model group,asiatic acid remarkably reduced brain water content,infract volume,and the concentrations of TNF-αand IL-1βin the brain tissue(P〈0.01or P〈0.05).Conclusion Asiatic acid provides significant protection against focal cerebral ischemia-reperfusion injury in rats.The protective mechanism is probably related to reduction of TNF-αand IL-1βlevel in the brain.
