鱿鱼硫酸软骨素糖蛋白的分离纯化和鉴定 被引量:15

Researches on Isolation, Purification and Identification of Chondroitin Sulfate-Glycoprotein from Sepia Illex Argentinu

摘要 目的:研究鱿鱼软骨硫酸软骨素的主要成分。方法:以鱿鱼喉软骨为材料,经提取、分离和纯化等步骤得到一酸性粘多糖组分C-S2;用高效凝胶过滤色谱和比旋光度鉴定其纯度,并用化学和光谱法研究其组成和结构。结果:C-S2为均一性较好的粘多糖,为硫酸软骨素C糖蛋白,其重均相对分子质量(Mw)149595,比旋光度-25.3。软骨素含量95.31%,蛋白含量2.59%,葡萄糖醛酸(GlcA)含量35.02%,己糖胺含量34.02%,总硫酸基含量9.54%,并存在少量中性单糖,由鼠李糖(Rha)、甘露糖(Man)、葡萄糖(Glu)和半乳糖(Gal)组成;β-消去反应说明C-S2中糖链以O-糖肽键连接到苏氨酸上。结论:鱿鱼软骨硫酸软骨素的主要成分为硫酸软骨素C糖蛋白。 Objective:To study the main composition of Chondroitin Sulfate from Sepia illex argentine. Methods:C-S2, an acidic mueopolysaccharide fraction was obtained from Ommastrephe bartrami by a series step of extraction, isolation and purification. The purity of C-S2 was proved by HPGFC and Polarimetry. Its composition and structure were studied by using chemical and spectral methods. Results: C-S2 was homogeneous and regarded as Chondroitin C glycoprotein with 2.59% protein, 95.31% ehondroitin content, 35.20% glucuronic acid, Hexosamine 34.02% and 9.54% sulfate content, its relative molecular weight and specific rotary were 149 595 and -25.3 respectively. A small quantity of neutral monosaccharide fraction existed were composed of rhamnose, mannose, glucose and galactose. β-elimination reactions showed that sugars and amino acids was linked by O-glycol-Ser bond. Conclusion:The main composition of Chondroitin Sulfate from Sepia iuex argentine was Chondroitin sulfate C glycoprotein.
