丝状真菌高分子量DNA的提取研究 被引量:15

摘要 为寻求一种简便高效的提取真菌基因组DNA的方法,采用经改良的液氮研磨饱和酚抽提法及高盐沉淀法,从液体培养的鸡土从菌菌丝体中提取了高分子量的基因组DNA。用紫外吸收光谱、限制酶切、RAPD—PCR反应及琼脂糖凝胶电泳等方法进行了鉴定。结果显示,两种方法均可以获得分子量大、样品较纯的基因组DNA,用限制酶均能有效地消化,并可有效地用于PCR扩增。其中高盐沉淀法提取的DNA产量高(每克菌丝体可获DNA0.83mg),方法简便,对人体无毒害。 In order to look for a facile and effcient protocol for extraction of fungal DNA, Genomic DNAs were extracted and isolated from fungal mycelia by two slightly modified methods: Liquid nitrogen grind ss phenol extraction and high concentration salt precipitation The properties of the DNAs were identified by U V absorption rate, restriction endonuclease digestion, RAPD PCR reaction and agarosegel electrophoresis The DNAs had a high molecular weight Tt was pure and thus is suited as substrate for restriction digestion and PCR The method of high concentrationsalt precipation is more convenient, efficient(DNA yield is 0 83mg/g mycelia) and safe Tt is more suitable for extraction of fungal DNA
