摘要
运用毛细管电泳-电化学检测法测定菊花、野菊与复方药中的5种主要营养成分,所选体系对木犀草素和槲皮素有很好的分离效果。研究了电极电位、缓冲液浓度和酸度、电泳电压和进样时间等实验参数对分离、检测的影响,得到了较优化的实验条件。以碳圆盘检测电极为工作电极,直径300μm,检测电位为0.95V(vs.SCE),在50mmol·L-1硼砂-硼酸(pH=9.2)缓冲液中,研究组分在20min内完全分离。5标样检出限(S/N=3)在0.9×10-6~2.5×10-6g·mL-1之间。
A high performance capillary electrophoresis (CE) with electrochemical detection (ED) method has been developed for determination of five active ingredients in Chrysanthemum morifolium Ramat,Flos Chrysanthemi Indici and its phytopharmaceutical products. The best separation parameters for luteolin and quercetin have been chosen. Some important factors such as the acidity and concentration of the running buffer, separation voltage, injection time and the potential applied to the working electrode are investigated for the optimum conditions. The working electrode is a 300 μm diameter carbon disc electrode positioned opposite the outlet of capillary. Under the optimum conditions, the analytes are separated within 20 min in a 70 cm length capillary at the separation voltage of 16 kV in a 50mmol.1^-l borate buffer(pH=9.2). Notably, excellent linearity is obtained over three orders of magnitude for all five analytes with detection limits (SIN=3) ranging from 0.9×10^-6 g.mL^-1 to 2.5×10^-6 g-mL^-1.
