摘要
通过分析比较禽流感病毒H5、H7和H9亚型不同毒株的基因组序列,设计出H5、H7和H9亚型特异性引物与探针,将扩增产物点制在芯片上,分别用H5、H7和H9亚型特异性探针与芯片杂交,研制出用于禽流感病毒H5、H7和H9亚型检测的正向杂交基因芯片,同时对正向基因芯片进行特异性、敏感性试验,并用该方法对待检样品进行了检测。结果表明,禽流感病毒正向基因芯片与新城疫病毒、传支病毒等6种禽类常见病毒无反应,其敏感性高于常规PCR方法。
Primers and probes were designed specific for Avian influenza viruses subtypes H5、H7 and H9 based on the genomic sequences that published at GenBank database.Amplified products of H5,H7 and H9 by PCR or RT-PCR were fixed on the slide,and hybridized with specific probes for subtypes H5,H7 and H9 separately.A microarray for detecting avian influenza viruses subtypes H5,H7 and H9 were developed.Then its specificity and sensitivity were validated by standard strains and samples from different areas.The results showed that the method based on microarray was of high specificity and sensitivity, and the method can meet the demands of field detection of AIV subtypes HS, H7 and H9 contamination.
