摘要
目的探讨不同剂量左旋多巴对形觉剥夺性弱视大鼠视觉诱发电位的影响及其可能的作用机制。方法30只14日龄SD大鼠幼鼠,随机分为3组,每组10只,分别为弱视对照组、小剂量给药组及大剂量给药组。3组SD大鼠采用单眼睑缝合30d建立形觉剥夺眼弱视模型。弱视对照组予以生理盐水灌胃,小剂量给药组与大剂量给药组分别予以20mg/kg、80mg/kg左旋多巴溶液灌胃给药。分别于给药前(45日龄)和给药后(75日龄)测量弱视大鼠模型的闪光视觉诱发电位(flash visual evoked potential,FVEP),并对所测得的数据进行统计学分析。结果给药前,各组剥夺眼较未剥夺眼FVEP的P1波潜伏期均明显延长(P<0.05)。给药后,小剂量组和大剂量组剥夺眼的P1波潜伏期较弱视对照组剥夺眼明显缩短(P<0.05),差异有统计学意义(P<0.05),且大剂量组较小剂量组P1波潜伏期缩短更明显(P<0.05);而各组未剥夺眼的P1波潜伏期比较,差异均无统计学意义(P>0.05)。各组剥夺眼与未剥夺眼N1P1和P1N2波振幅比较,差异均无统计学意义(P>0.05)。结论左旋多巴能缩短弱视鼠弱视眼的P1波潜伏期,而对振幅N1P1和P1N2无明显作用。它可能通过改变视网膜内及整个视路的多巴胺含量而影响视功能。
Objective To study the effect of different doses of levodopa on the flash visual evoked potential (FVEP) of formdeprived amblyopic rats, and to discuss the possible mechanism of amblyopic therapy. Methods Thirty rats were randomly divided into 3 groups, each consisting of 10 rats: an amblyopia control group, a low-dose levodopa group (LD, 20 mg/kg), and a high-dose levodopa group (LD, 80 mg/kg). Form-deprived amblyopia rat models were established by suturing the eyelids of the test eye for 30 days, then FVEPs were measured before the administration of levodopa (at 45 days of age) and after administration (at 75 days of age). Results The latent periods of the P1 waves of form-deprived eyes in each group were significantly longer compared to non-deprived eyes before administration (P〈0.05). The latent periods of the P1 waves of form-deprived eyes in both the high- and low-dose levedopa groups were significantly shorter than those of the amblyopic control group (P〈0.05) after administration. In addition, the latent periods of the P1 waves for the high-close levodopa group were shorter than those of the low-dose group (P〈0.05). However, there was no significant difference between the latent periods of the P1 waves in the non-deprived eyes for all groups, and the amplitudes of waves N1P1 and P1N2 were only slightly altered (P〉0.05). Conclusion Levodopa can decrease the latent period of the P1 wave in amblyopie eyes, improving visual function in amblyopic eyes, and thus influencing the sensitive period for plasticity in the development of the visual system.
