大鼠骨髓基质干细胞在与成骨细胞共育环境下的成骨特性

摘要 目的在与成骨细胞(OB)共培养的条件下,观察大鼠骨髓基质干细胞(MSCs)的成骨特性。方法取自3月龄大鼠骨髓分离MSCs,扩增后以免疫组化法鉴定其表型,OB取自1日龄乳鼠,酶消化法获得。传代后的MSCs消化后分为MSCs/OB共培养组与普通培养组,观察MSCs的形态学变化及矿化能力,20 d后半定量RT-PCR方法检测两组骨钙素的表达,茜素红染色后显微镜下计数钙化结节。结果共培养组的MSCs形成的钙化结节数高于普通培养组(P<0.01);共培养组的MSCs,其细胞内骨钙素表达量明显高于普通培养组(P<0.05)。结论与OB共培养的MSCs成骨能力增强。
