缺血后处理及腺苷后处理对大鼠肺缺血再灌注损伤的作用 被引量:1

Effects of ischemic postconditioning and adenosine postconditioning on lung ischemia-reperfusion injury in rats

摘要 目的探讨缺血后处理及腺苷后处理对大鼠肺缺血再灌注损伤的作用。方法通过阻断右肺门建立大鼠在体右肺缺血再灌注模型,40只健康的SD大鼠随机分为4组,每组10只。(1)假手术组:持续灌注165min;(2)缺血再灌注组:缺血45min,再灌注120min;(3)腺苷后处理组:缺血45min后,予腺苷1.5mg·kg-1右心室弹丸注射,然后再灌注120min;(4)缺血后处理组:缺血45min后,短暂再灌注10s,缺血10s,反复5次,然后再全面恢复灌注120min。实验结束时取各组肺组织测湿/干重(W/D)值,HE染色行组织学检查,ELISA测定IL-6、IL-10含量,紫外分光光度计测定丙二醛(MDA)、超氧化物歧化酶(SOD)含量。结果缺血后处理组与腺苷后处理组对大鼠肺缺血再灌注损伤的保护作用类似。与缺血再灌注组相比较,缺血后处理组与腺苷后处理组W/D显著降低(均P<0.05),SOD含量显著升高(P<0.01)而MDA显著降低(P<0.05),IL-6含量有所降低而IL-10含量有所升高(均P<0.01),光镜下肺损伤亦有所减轻。结论缺血后处理和腺苷后处理可能通过脂质抗氧化作用,灭活氧自由基,抑制致炎细胞因子IL-6,上调保护性细胞因子IL-10,从而减轻肺缺血再灌注损伤。 Objective To study the effects of ischemic postconditioning and adenosine postconditioning on lung ischemia-reperfusion injury in rats.Methods Rat models of right lung reperfusion iniury were induced by occluding hilum of right lung.Forty SD rates were randomly divided into four groups:sham operated group(n=10):continuous perfusion for 165 minutes;ischemic-reperfusion group(n=10):ischemia for 45 minutes and reperfusion for 120 minutes;adenosine postconditioning group(n=10):ischemia for 45 minutes,adenosine(1.5 mg·kg-1) bolus injection through right ventricle before reperfusion,then reperfusion for 120 minutes;ischemic postconditioning group(n=10):ischemia for 45 minutes,transient reperfusion for 10 seconds,ischemia for 10 seconds,which were repeated for 5 times,and then recovered to comprehensive reperfusion for 120 minutes.In the end of experiment,the lung tissue was taken to determine the wet/dry (W/D);the lung tissue structure was observed by using optics-scope.ELASA was used to determine the contents of IL-6 and IL-10.UV spectrophotometer was used to determine the content of MDA and SOD.Results Compared with ischemia-reperfusion group,ischemic postconditioning group and adenosine postconditioning group had similar effects:the wet-to-dry ratio decreased significantly(P〈0.05);content of SOD increased significantly(P〈0.01) and MDA decreased significantly(P〈0.05);content of IL-6 decreased and IL-10 increased(all P〈0.01).Conclusions Ischemic postconditioning and adenosine postconditioning can provide protective effects on lung ischemia-reperfusion injury by preventing lipid peroxidation and free radical formation,inhibiting proinflammatory cytokines IL-6 and increasing protective cytokine IL-10.
