正交实验法优选盐酸川芎嗪的超声提取工艺 被引量:2

摘要 目的优选川芎中盐酸川芎嗪的最佳超声提取工艺。方法采用正交实验法,用高效液相色谱法进行测定,以盐酸川芎嗪为指标,考察溶剂用量、乙醇体积分数、超声时间等因素对川芎提取效果的影响。结果实验设计的3因素中对结果影响最显著的因素为超声时间。盐酸川芎嗪的最佳提取工艺为:5 g川芎粗粉,30%的乙醇(含HCl 0.1 mol.L-1)50ml,超声60 min。最佳条件下盐酸川芎嗪得率为0.560 4 mg/g。结论该法简便、快速、节能、溶剂用量少、提取效率高,适宜于川芎中川芎嗪的提取。 Objective To optimize the ultrasonic extraction process for ligustrazine hydrochloride from Ligusticum chuanxiong Hort. Methods Using orthogonal design, the effect of solvent consumption , alcohol volume fraction and ultrasonic time on the extraction results were observed , with the content of Ligustrazine hydrochloride as the index by high performance liquid chromatography. Results Among the factors of test, the most significant effect was the ultrasonic time. The optimal conditions for the extraction of Ligustrazine hydrochloride were as follows: adding 50 ml of 30% alcohol( include 0.1 mol.L^-1 HCl ) to the herbs (5 g), and extracting for 60 minutes by uhrasonic (ultrasonic power was 100 W). The extract yield was 0. 5604mg/g . Conclusion Because of its simplicity , rapidness, low solvent consumption and high efficiency of extraction , the method of ultrasonic extraction can be successfully used in extraction for Ligustrazine hydrochloride from Ligusticum chuanxiong Hort.
