摘要
采用田菁固氮根瘤菌AzorhizobiumcaulinodansORS571(pXLGD4)加或不加类黄酮侵染8个甘蓝型油菜品种,即高硫甙葡萄糖甙(硫甙)、高芥酸,高硫甙、低芥酸,低硫甙、高芥酸,低硫甙、低芥酸4种类型的品种。所用菌株以LacZ为报告基因,侵染部位为侧根缝隙。结果表明,类黄酮能有效地促进高硫甙品种的侵染率。通过切片和显微镜观察,细菌存在于根内外皮层细胞间隙并能在细胞间隙移动。从油菜根系分离出的细菌回接到原寄主(田菁)上能形成有效根瘤。
The abilities of Azorhizobium caulinodans ORS571(pXLGD4) to infect 8 varieties of Brassica napus have been studied, the varieties were high glucosinolates, high erucic acid; high glucosinolates, low erucic acid; low glucosinolates, high erucic acid and low glucosinolates, low erucic acid. Using strain tagged with LacZ reporter genes, the principal colonization site was found to be the points of emergence of lateral root cracks(LRCs). The results showed that flavonoids could improve LRCs(%) in varieties with high glucosinolate content. Minute structure of root sections showed clearly that rhizobia were existed interal of cells, and distributed from outer corter to inner corter. A low nitrogenase activity was determined by the method of acetylene reduction. The strain were isolated from root of rapeseed and successfully reinoculated to original host.
