人神经营养素-3在昆虫杆状病毒表达系统中的表达 被引量:1

摘要 将人NT 3基因插入 pPAK9中后与Bsu36Ⅰ酶切的Bac6昆虫杆状病毒基因组DNA共转染sf2 1细胞 ,以空斑法分离重组病毒 .狭缝杂交、细胞免疫化学染色和背根节无血清培养证明NT_3整合入Bac6DNA中 ,表达并分泌至胞外 .2 0 0mL培养上清经CMSepharoseFastFlow纯化得到 2 0 0 μgNT_3纯品 ,其最大生物效应浓度为 4ng/mL .
