摘要
烷基酚作为一类重要的环境污染物已经对男性生殖产生很大的负面影响.本文的主要目的是对比研究不同烷基酚对雄性大鼠睾丸支持细胞的影响.各种烷基酚,包括甲基酚,丁基酚和壬基酚处理支持细胞后,对比研究它们对支持细胞膜通透性和细胞外基质产生的影响.实验结果表明各类烷基酚在处理12 h内均能诱导细胞膜通透性(以lactate dehydrogenase(LDH)泄漏率表示)增加,并具有一定的剂量依赖效应,其中丁基酚和壬基酚的作用强于甲基酚;当处理时间增加至24 h时,各组LDH的泄漏率均在较低浓度即达到最大(丁基酚和壬基酚5×10-8mol/L,甲基酚10-7mol/L)并不再随浓度增加而增大.此外,各种烷基酚处理均能增加支持细胞层粘连蛋白产生量,并具有一定剂量依赖性,其中丁基酚的刺激作用强于甲基酚和壬基酚.
Alkylphenols(APs) as important environment contaminants have caused disruptive effects on male reproduction.The aim of present study was to compare different alkylphenol-induced effect on rat testicular sertoli cells.Alkylphenols,including p-cresol,4-n-butylphenol and 4-n-nonylphenol,were treated to purify rat testicular sertoli cells,and their effects on cell membrane permeability and extracellular matrix production were studied and made comparison.The results revealed that alkylphenols increased membrane permeability(indicated as lactate dehydrogenase(LDH) leakage ratio) of sertoli cells in a dose-dependent manner within 12h treatment,and the effect of butylphenol and nonylphenol was stronger than that of cresol which holds a shorter alkyl chain than the other two alkylphenols.When exposure time was extended to 24h,LDH leakage reached a high-point at very low concentrations(5×10-8mol/L for butylphenol and nonylphenol,10-7mol/L for cresol) and did not increase with dosage.In addition,exposure to alkylphenols induced laminin production in a dose-dependent manner,and the stimulative effect of butyphenol was more effective than that of cresol and nonylphenol.
