硫酸庆大霉素对小鼠耳蜗螺旋神经节细胞生长的影响 被引量:7

摘要 为观察硫酸庆大霉素对培养的小鼠耳蜗螺旋神经节细胞(SGC)的细胞生存数和细胞突起长度的影响,在单离培养的出生1~5d的NIH小鼠SGC培养液中,加入不同浓度的硫酸庆大霉素(25,50,100mg/L)。培养14d后,标本用4%多聚甲醛固定,甲苯胺蓝染色,在光镜下计数SGC生存数,目镜测微尺下测量细胞突起长度。结果显示,实验组与对照组间、不同浓度实验组间的细胞生存数及细胞突起长度无明显差异(P>0.05)。提示硫酸庆大霉素对小鼠耳蜗SGC的生存和生长无明显影响,氨基糖甙类抗生素对SGC的影响可能是继发性的。 The effect of gentamicin on survival and neuritogenesis of cultured spiral ganglion cell(SGC) from NIH mouse 1-5 days after birth was observed. Different doses (25,50 and 100 mg/ L )of gentamicin were add to the medium of SGC,and cultured for 14 days. After that,the specimen was fixed with 4% paraformaldehyde and stained with toluidine blue. The amount of survival SGC was counted,and the length of neurite was mesured with micrometer under microscope. It was shown that the survival and length of neurite of SGC had no difference between Gentaforcin treated groups and control groups,and there were also no difference among 3 gentamicin treated groups. It was suggested that gentamicin had no effect on survival and neuritogenesis of SGC.The effects of aminoglycoside antibiotics on SGC were probably secondary to the injury of Corti's Organ.
