摘要
串联亲和纯化(tandem affinity purification,TAP)最早作为一种研究酵母细胞内天然状态下蛋白质相互作用的工具,目前不仅在多个技术领域已成为筛选鉴定新蛋白质复合体的重要工具,并且将成为系统生物学研究的一项关键技术,在探索和绘制生理条件下多细胞生物蛋白质相互作用网络图谱中发挥主导作用。本文就TAP近些年来的研究进展进行了概括,并对其常见问题与改进策略进行了总结。TAP技术的不断完善,必将使其在蛋白质相互作用领域发挥更重要的作用。
Tandem affinity purification (TAP), initially used as an approach to study in vivo protein interactions of yeast under natural state, is now extending to many other fields. At present, TAP has become an important tool to identify as well as purify new protein complexes, and will hopefully become a key supporting technology in system biology, playing its role in system- atically mapping in vivo multi-protein complexes. But despite its incomparable advantages and wide prospect of application, some problems exist as well. In this paper, problems emerging from the application of TAP in recent years and tag improvement for different purposes were reviewed. Hopefully, a more efficient and systematic strategy for improving TAP tags will be developed soon to promote the application of TAP in related fields.
