摘要
实验研制了胰岛素滴眼剂。通过对缓冲液、增粘剂和吸收促进剂的筛选,确定了最佳处方组成:2%胰岛素、1%Brij-78、0.5%EDTA、1%玻璃酸钠的含0.03%对羟基苯甲酸乙酯的硼酸缓冲液。对兔眼试验证明该制剂无刺激性。药效学实验表明用其滴眼后吸收迅速,可显著降低糖尿病兔的血糖。血糖下降持续时间和最大降血糖均与给药剂量呈正相关。
Insulin eyedrops were studied by selecting buffer system, viscosity improver and absorption promoters, and the optimal composition of the preparation, which was a borate buffer containing 0.03% ethylparaben, 2 %insulin, 0.5% EDTA, 1% Brij-78 and 1 % sodium hyaluronate, was determined. It was proved that the preparation had no irritution to rabbit eyes. The results of pharmacodynamic experiment showed that insulin was rapidly absorpted and the plasma glucose concentration reduced significantly after ocular instillation to diabetic rabbitS. There were positive relationships between dosages and the duration of the lower blood glucose, and the maximum reduction rate of blood glucose levels.
