摘要
对280株不同种属的酵母菌株进行了初筛、复筛、单倍体分离、诱变和杂交,从杂交菌株中选育到1株高生物量富铬酵母菌株(编号为ZGH-8)。并初步优化了发酵条件:培养基为2%糖浓度的蔗糖糖蜜、0.5%酵母粉、0.5%尿素、0.1%H3PO4、铬添加量800μg/mL,pH6.0,装液量50mL/250mL三角瓶,培养时间30h。在优化条件下,杂交菌株ZGH-8的生物量(细胞干重)可达到7.6g/L,细胞铬含量可达3100μg/g。
High-biomass, chromium-enriched yeast strain ZGH -8 was constructed by primary screening, isolation of haploid, mutagenesis and hybridization. The primary optimization of cultivation conditions was determined through fermentation tests. The highest biomass and Cr content was obtained when strain ZGH - 8 was cultured for 30 h in 50 mL of molasses medium in 250 mL shake flasks containing 2% sugar, 0.5% yeast extract, 0.1% HaPO4 and 800μg/mL Cr. The initial pH was adjusted to 6.0. Under the optimized cultivation conditions, the biomass( dry weight) reached 7.6g/L and the Cr content reached 3 100 μg/g.
