摘要
以苦瓜为原料,建立了SPE精制、HPLC检测、ESI-MS定性为主要步骤的苦瓜皂苷的分析方法。通过HPLC测定,得到了SPE精制的最佳工艺条件为:上样pH为8,经10%、20%乙醇各15mL淋洗后用10mL40%乙醇淋洗,洗脱液为65%乙醇,洗脱体积为7mL,流速为1mL/min,此条件下所得液相色谱的峰数、峰形、峰面积均较理想。测得最佳条件下精制的苦瓜总皂苷含量为82.13%。经ESI-MS定性分析得出,精制后的产物为三萜类皂苷。苦瓜皂苷快速分离方法的研究,对其构效关系和生物活性研究提供了重要的前提条件。
The analysis method of momordica saponins with SPE purification, HPLC detection and ESI-MS qualitative analysis was established using momordica as raw material. The optimum conditions of SPE purification were determined by HPLC, and the results were as follows: the sample pH value was 8% it was leached with 10 mL 40% ethanol after washing with 10% and 20% ethanol; the elution solution was 7 mL 65% ethanols and the flow rate was 1 mL/min. On this condition, the peak numbers, peak shape and peak areas of the HPLC profile were all relatively ideal. Under optimum conditions, the content of momordica saponins was 82.13 %. The ESI-MS qualitative analysis result showed that the refining product was triterpenes saponins. The rapid separation method of momordica saponins proindes the basis for the study of their structure-effect relationship and biological activity.
