摘要
目的为乳腺癌诊断提供血清学新标志。方法免疫PCR方法检测血清抗p185蛋白抗体，酶免疫组化方法检测组织p185蛋白表达。结果乳腺癌患者血清抗p185蛋白抗体阳性率为30％，而非癌患者和正常人血清抗p185蛋白抗体均为阴性，乳腺癌患者血清中抗p185蛋白抗体显著高于非癌患者和正常人（P〈O．01）。p185蛋白阳性表达的乳腺癌患者抗p185蛋白抗体阳性率为69．2％，明显高于p185蛋白阴性表达组，血清p185抗体测定与p185蛋白表达密切相关（P〈0．01）。结论检测血清抗p185蛋白抗体是检测组织p185蛋白理想的替代工具。抗p53蛋白抗体可以作为乳腺癌血清学诊断新标志，用于乳腺癌的普查和早期诊断。
Objective To establish a new immuno-PCR assay for detecting seral anti-p185 antibodies,to investigate the re- lation of anti-p185 antibodies in patients with breast carcinoma to 185 protein expressions in tissue and clinical signifi- cances. And to afford a practical tool in diagnosis of breast carcinoma clinically. Methods Anti-p185 antibodies were de- tected by immuno-PCR assay in sera of patients with breast carcinoma,noncarcinoma and health controls. P185 protein ex- pressions were detected by immunohistochemistry assay. The relation of anti-p185 antibodies in sera to p185 protein ex- pressions in tissue were analyzed. Results The positive rate of anti-p185 antibodies by immuno-PCR was 30% in 60 sera of patients with breast carcinoma. None of anti-p185 antibody was found in 30 nonearcinoma and 60 health controls. The statistics results indicated the positive rate of anti-p185 antibodies was significantly higher in patients with breast carcino- ma by immuno-PCR than noncarcinoma and health controls（P^0. 01） and there was no difference between patients with noncarcinoma and health controls （P^0. 05）. The specificity and the sensitivity of immuno-PCR for anti-p185 antibody were 100% and 30%. The positive rate of anti-p185 antibodies was 69.2~~ in 26 cases of sera of breast carcinoma with positive p185 protein expressions and was significantly higher than that of negative p185 protein expressions（P-~0. 01）. Conclusion Anti-p185 antibodies in sera are positively corralation to p185 protein expressions in tissues. Immuno-PCR for anti-p185 antibodies in sera assay is simpler than immunohistochemistry assay for p185 protein expressions in tissue and therefore is a practical tool.
