摘要
采用正交试验法探讨枸骨叶抑菌物质的最佳提取条件,以杯碟法对枸骨叶提取物进行体外抗菌作用以及pH、温度、时间因素对其抗菌活性的影响。结果表明:枸骨叶抑菌物质的最佳提取条件为:以乙酸乙酯为溶剂,提取温度为40C,料液比为1:10,浸提时间为2.5h;枸骨叶提取物对金色葡萄球菌、枯草杆菌、大肠杆菌都有明显的抑菌效果,最低抑菌浓度分别为0.0625、0.5、0.5mg/mL;最小杀菌浓度为0.125、1、0.5mg/mL。该物质抑菌pH范围为7~9,热稳定性较差,最佳抑菌时间为24h。
The optimum extractive conditions of bacteriostasis from Folium Ilicis Cornutae were studied by orthogonal experiment, then the bacteriostatic activity of Folium Ilicis Cornutae extracts and the effects of temperature,time,pH value on it were studied by cylinder plate method. The results showed the optimum extractive conditions of bacteriostasis from Folium Ilicis Cornutae was: ethyl acetate solvent,leaching-temperature 40℃ ,ratio of solid to liqnid 1 : 10, extracting time 2. 5 h. The substance extracted showed some antibiotic activity against Staphylococcus aureus, Bacillus subtilis and Escherichia coli, the minimum inhibitory concentration(MIC)was 0. 0625,0. 5,0. 5 mg/mL, and minimum bactericidal concentrations(MBC)was 0. 125,1,0. 5 mg/mL respectively. The antibacterial effects was best at pH 7-9. The antibacterial activity of Folium Ilicis Cornntae extracts was unstable under heating,The optimum antibacterial time was 24 h.
