摘要
目的:建立血浆中咪达唑仑浓度的高效液相色谱-紫外检测方法。方法:采用inertsilODS色谱柱;流动相为甲醇-水(62:38),水相含磷酸0.16%,二乙胺0.25%;流速为1mL.min-1。采用紫外检测,检测波长为225nm。结果:咪达唑仑保留时间分别为12.86min、19.15min,咪达唑仑在0.4~12.8μg.mL-1范围内线性关系良好,回归方程为:Y=15448X+313.47,r=0.9993;日内及日间变异均<5%。结论:用高效液相色谱-紫外法检测咪达唑仑的血药浓度,方法简便、稳定,可满足临床研究的需要。
OBJECTIVE:To establish an HPLC method for the determination of plasma concentration of midazolam.METHODS:The samples were separated on inertsil ODS column with mobile phase consisted of Methanol-water(62:38,V/V).The water phase contained 0.16% phosphoric acid and 0.25% diethyl amine.The UV detection was performed under a detection wavelength of 225 mn.RESULTS:The retention time of midazolam was 12.86 min.The linear range of midazolam was 0.4~12.8 μg·mL-1(r= 0.999 3).The regression equation was expressed as Y= 15 448 X+313.47(r= 0.999 3).Both the intra-day and inter-day coefficients of variation were less than 5%.CONCLUSION:This HPLC-UV is simple and stable for the determination of plasma concentration of midazolam,and it meets the requirement on clinical research.
