摘要
采用高速逆流色谱法分离纯化玫烟色拟青霉菌丝体中的过氧麦角甾醇。溶剂系统为V(正己烷):V(乙腈):V(三氯甲烷)=5:5:1,上相为固定相,下相为流动相,转速为850r/min,流速为2mL/min。所得产物纯度经高效液相色谱分析为98.5%(峰面积归一化法),经高分辨ESI-MS质谱确认其精确分子量。
High-speed-counter-current chromatography(HSCCC) was applied to the separation of peroxy-ergosterol from a mycelia strain of Paecilomyces fumosoroseus. The solvent system was n-hexane-acetonitrile-chloroform (5 :5 : 1 ,v/v/v) ; the upper column was solid phase and the lower column was the mobile phase with the flow rate of 2.0 mL/min and rotation speed at 850 r/min. The obtained fraction was determined by HPLC with 98.5% purity. The molecular formula of the compound was identified by ESI-MS.
