摘要
[目的]为中药大黄的快速定性分析提供技术支持。[方法]以青海大黄为试材,制备不同浓度的大黄水浸液并对其进行荧光扫描;取一定浓度的大黄水浸液,加入不同pH值的HOAc—NaOAc缓冲溶液后对其进行荧光扫描。[结果]随着大黄水浸液浓度的减小,荧光强度逐渐减小,其荧光强度与浓度呈线性关系。当缓冲溶液的pH值为2.6~4.2时,荧光强度随着pH值的增大而增大。当缓冲溶液的pH值为4.2—5.8时,荧光强度随着pH值的增大而减小。当缓冲溶液的pH值为4.8~5.2时,荧光强度几乎不变。中药大黄水浸液的三维荧光等高线光谱图中有一个明显的荧光峰,其发射波长位于401.8nm。[结论]该试验确定了中药大黄水浸液荧光光谱分析法的测定条件。为中药大黄的定性分析提供了新方法。
[ Objective ] The purpose of the study was to supply technical support for the rapid qualitative analysis on Chinese medicine rheum. [ Method] The rheum aqueous extracts with different concn, were prepared with Qinghai rheum as tested material and the fluorescence scan- ning was performed on the aqueous extracts. The fluorescence scanning was performed again after HOAc-NaOAc buffer solutions with different pH values were added into the selected rheum aqueous extracts with certain conch. [ Resuh ] As the concn, of rheum aqueous extracts was re- duced, the fluorescence intensity was reduced gradually, its fluorescence intensity showed a linear relationship with its conch. When the pH value of buffer solution was 2.6 - 4.2, the fluorescence intensity was increased as the pH value was increased. When the pH value of buffer solution was 4.2 - 5.8, the fluorescence intensity was reduced as the pH value was increased. When the pH value of buffer solution was 4.8 - 5.2, the fluorescence intensity was nearly invariable. There was an obvious fluorescence peak in the three-dimensional fluorescence contour spectrogram of the aqueous extracts of Chinese medicine rheum and its emission wavelength was at 401.8 nm. [ Conclusion] The condition of determining the aqueous extracts of Chinese medicine rheum by fluorescence spectrum analysis was ascertained in this experiment. It provided a new method for the qualitative analysis on Chinese medicine rheum.
