摘要
目的观察成年期甲状腺功能减退症(简称甲减)大鼠海马神经突触前膜胞内蛋白Munc18表达及常规剂量和大剂量替代治疗后的变化。方法用丙基硫氧嘧啶(PTU)建立成年期大鼠甲减及治疗模型,放射免疫法测定血清甲状腺素水平,免疫组织化学法观察Munc18在大鼠海马结构的表达与分布。结果与正常对照组比较,甲减大鼠血清T3、T4水平减低,差异有统计学意义,常规剂量替代治疗后恢复正常,大剂量替代治疗后升高,差异有统计学意义。免疫组织化学法显示甲减组海马CA3区、齿状回(DG)和CA1区多形层的Munc18平均光密度值减低,常规剂量替代治疗后,仅DG区分子层平均光密度值减低,大剂量替代治疗后,其差异无统计学意义。结论成年期甲减可致海马内Munc18蛋白表达减少,常规剂量甲状腺素替代治疗能使其大部分恢复,大剂量替代治疗能全部恢复。
Objective To observe the expression of Mncl8 in hippocampus of the adult rat with hypothyroidism and change after regular-dose and high-dose treatment therapy with thyroid hormone. Methods The adult male SD rats were replicated to the adult-onset hypothyroidism and treatment models with propyhhiouracil (PTU). We used ra- dioimmunity method to assay the levels of serum thyroxine, immunohistochemical technology to assay the levels of Munc18 protein in different subfields in hippocampus. Results Compared with the normal control group, the levels of serum T3 and T4 were lower disparity have statistical significance in the control hypothyroidism group(CH), similar in the L-T4 treatment group with regular dose (TYR) , and higher disparity had statistical significance in the L-T4 treatment group with high dose(TIM). The levels of Munc18 protein in the CA3 ,polymorphic layer of CA1 and DG regions of hippocampus were lower in the CH group, only in the molecular layer of DG was lower in the TTR group, and disparity had no statistical significance in the TTH group. Conclusion Adult-onset hypothyroidism eause the levels of Munc18 decrease in dorsal hippoeampus, the majority levels of Munc18 protein return to the normal by regular-dose replacement and the all levels of Munc18 protein return to the normal by high-dose treatment.
