摘要
为组织培养快速繁殖云南独蒜兰,对其进行了种子无菌萌发的基本培养基、添加物与激素以及原球茎增殖激素配比的优化筛选。结果表明:在KC、1/2MS、B5这3种基本培养基中,B5较适于种子萌发;添加蛋白胨5 g/L、马铃薯泥50 g/L或活性炭粉1 g/L均有利于种子萌发形成原球茎;生长激素NAA、2,4-D对种子无菌萌发具有促进作用,而细胞分裂素6-BA则抑制了种子萌发。在以B5附加AC(活性炭)1 g/L的基础培养基中,激素组合6-BA 1.5 mg/L+NAA 0.5 mg/L更有利于原球茎的增殖与生长。
Optimial screening of basic media, additives and plant hormones for aseptic seeding and combinations for protocorm-like body multiplication was studied. The aseptic seeding of Pleione yunnanensis showed that among KC,B5 and 1/ 2 MS, B5 was the optimum basic medium for germination. Supplement of protein peptone 5 g/L, potato 50 g/L or AC (active carbon) 1 g/L was advantageous to seed germination and PLB induction. NAA and 2, 4-D showed positive effects on germination, while 6-BA might play a suppressive role. On the medium of B5 supplemented with AC, 6-BA 1.5 mg/L+NAA 0.5 mg/L was the best combination for PLB multiplication and its growth.
