摘要
目的建立栀子中京尼平苷的含量测定方法,在此基础上考察栀子中京尼平苷的提取方案。方法高效液相色谱法测定含量,采用KromasilC18(250mm×4.6mm,5μm)色谱柱,流动相为乙腈-水(15∶85);流速为1.0ml/min;检测波长为238.4nm。栀子粉末先醋酸乙酯回流提取,滤渣再用80%乙醇回流,考察提取效率及京尼平苷纯度。结果京尼平苷在12.5~200μg/ml范围内呈良好线性关系,r=0.9998。用乙醇回流的提取方式可使京尼平苷提取较完全,醋酸乙酯提取所得京尼平苷纯度较高。结论提取溶剂的种类对栀子中京尼平苷的提取效率及纯度有较大影响,先醋酸乙酯再乙醇的提取方法,可以得到部分纯度较高的京尼平苷,且不浪费药材,减少纯化等后续处理的难度。高效液相色谱法检测栀子中京尼平苷,方法快速、准确、可行。
Objective To develop an HPLC method for the determination of geniposide from gardenia. Methods High performance liquid chromatography was used on ODS C18 (250 mm×4.6 mm,5 μm)with acetonitrile- water (15: 85 ) as a mobile phase. The flow rate was 1.0 mL/min. The detection wavelength was 238.4nm. First a circumfluence was made by ethyl ,the residue was extraced by 80% ethanol. Then the rate and purity of Geniposide extracted via different extraction solvent and extraction method. Results The linear range of geniposide was 12.5 -200 μg/ml, r = 0. 999 8. The extraction method of ethanol could get a higher extraction rate of Geniposide, and a high purity with ethyl acetate instead. Conclusion The type of solvent in the extraction of Geniposide from Gardenia affects the efficiency of extraction significantly. Make a sequential extraction by using ethyl first and then the ethanol, we can not only get the Geniposide in a high purity, but also waste little herbs. Determination of Geniposide in Gardenia can be done fast and exactly by HPLC .
