A Simple Model Establishment Method for Rat Delayed Cerebral Vasospasm after Subarachnoid Hemorrhage

摘要 目的寻找一种简单、有效、易重复的大鼠蛛网膜下腔出血(SAH)后迟发性脑血管痉挛(CVS)的模型。方法将18只SD大鼠随机分为3组,每组6只。SAH组:经枕大池穿刺注射自体股动脉血200μL制作SAH模型,24 h后重复;非SAH组:以等量的0.9%氯化钠注射液行枕大池注射,24 h后重复;正常对照组:不进行任何处理。观察2次枕大池穿刺后5 d的临床表现,基底动脉的直径、横切面积变化及形态学的改变。结果与正常对照组比较,SAH组大鼠精神萎靡、嗜睡、活动减少,蛛网膜下腔均可见弥漫性分布的血液或血凝块,基底动脉直径、横切面积明显变小(P<0.001),形态学观察改变明显;非SAH组则无明显改变。结论本模型操作稳定可靠、简单、易重复,适合进行SAH后迟发性CVS的相关研究。 Objective To find a simple,effective and easily repeatable model of delayed cerebral vasospasm (CVS) after the rat subarachnoid hemorrhage (SAH). Methods 18 SD rats were randomly divided into 3 groups.SAH group (n=6) ,non-SAH group (n=6) ,and the normal control group (n = 6). For SAH group,autologous arterial blood (200 μL)was injected into cerebellomedullary cistern of the rats,which was repeated once 24 h after. For non-SAH group, equivalent amount of 0.9% sodium chloride solution instead of autologous blood was injected into cerebellomedullary cistern,which was also repeated once 24 h after. For the control group,no treatment was made. 5 days after the the second cerebellomedullary cistern injection,observation was made on the behavior,diameter and transverse area of the basilar artery,and morphological change. Resuits Compared with the normal control group,the rats in SAH group were apathetic, drowsy, inactivitive,and diffuse distribution of blood or blood clot in subarabchnoid could be visible. For S,h,H group, the diameter and transverse area of basilar artery turned out significantly smaller than the control group(P〈0.001) ,and the morphological change was significant,while there was notsignificant change in non-SAH group. Conclusion The model establishment method is stable, reliable,simple and easily repeatable,which is suitable for study of delayed CVS after SAH.
