摘要
[目的]探寻有效的轮叶黑藻芽孢预处理技术。[方法]采用80μW/cm2紫外光照射、40 kHz超声波处理2种方式对轮叶黑藻芽孢进行预处理,并观察其在TP浓度为0.1 mol/L、TN浓度为4.0 mol/L的富营养化水体中的萌发、生长情况。[结果]结果表明,紫外预处理能够提高轮叶黑藻芽孢过氧化物酶活性和可溶性糖含量;超声预处理后,过氧化物酶活性提高,而可溶性糖含量略有降低。随着芽孢生长,2组轮叶黑藻的过氧化物酶活性均略低于对照;而可溶性糖含量与对照组差异不大,都呈现先升高后降低的趋势。2种预处理方式下轮叶黑藻的生长指标发生了相应的变化,紫外组与对照组相比,生长25 d后其生物量增加0.18 g/ind.,植株高度增加4 cm,平均分枝数增加0.33个;超声组与对照组相比,只有发芽率增长比较显著,从83%提高到100%。[结论]紫外预处理能在一定程度上促进轮叶黑藻芽孢生长。
[ Objective] The purpose of this research was to discuss the effective pretreatment technology for Hydrilla verticillata propagule. [ Method ] The sprouting and growth of H. verticillata propagule in eutrophic water of 0.1 mol/L TP and 4.0 mol/L TN were investigated after pretreatments in two ways: treated by 80 μW/cm^2 UV-light and a frequency of 40 kHz for 5 min. [ Result ] The results showed that : the peroxidase activity and the soluble carbohydrate content were both increased after the propagules were pretreated by UV radiation, and the peroxidase activity was increased while the soluble carbohydrate content was decreased slightly after the propagules were pretreated by ultrasound. As the propagules grew up, the peroxidase activities of the two groups were lower than that of the control; the soluble carbohydrate contents of the two groups increased at first and then decreased. The indices of growth were changed after the pretreatments. As for the groups pretreated by UV radiation, the biomass, the height of plants and the number of branches increased by 18 g/ind. , 4 cm and 0.33 per one in 25 days of growing, respectively. Only the sprouting rate increased from 83% to 100% after pretreatment by ultrasound. [ Conclusion] The pretreatment with UV radiation to propagules can accelerate the growth of H. verticillata obviously.
