摘要
目的探讨力竭游泳运动对大鼠血管平滑肌细胞中间纤维和超微结构的影响,为运动对血管的生理和病理变化研究提供一定的形态学依据。方法SD大鼠进行1次力竭游泳(SE)和1周力竭游泳(SCDE),分别在1次或末次力竭游泳运动后即刻(0 h)、3 h2、4 h、48 h和100 h,取胸主动脉平滑肌分别进行石蜡切片样本处理和常规透射电镜样本处理,石蜡切片分别进行结蛋白和波形蛋白的免疫组织化学染色(Envision法),观察各组结蛋白和波形蛋白的分布,计算其平均光密度和阳性面积的改变,电镜观察肌细胞内超微结构的改变。结果1次力竭运动后血管平滑肌的结蛋白、波形蛋白和超微结构与对照组相比,未见明显改变;经过1周力竭运动训练后,平滑肌的结蛋白和波形蛋白表达降低,超微结构可见损伤性改变。结论长期力竭游泳运动对大鼠血管平滑肌中间纤维和超微结构有显著的影响,并且在运动后不能短期内恢复。
Objective To explore the effects of single exhaustive swimming on median fiber and ultrastructure of rats' VSMC. so as to provide some morphological basis of the changes of physiology and pathology by the exerciseinduced change of VSMC. Methods After exercises of SE and SCDE, rats' smooth muscles were cut off on 0 h, 3 h, 24 h, 48 h and 100 h. These specimens were treated respectively with regular procedures of paraffin sections and transmission electron microscope (TEM). The paraffin sections were stained by Des and Vim immunhistochemistry, their mean optics density and percentage of stained areas were observed and calculated. The ultrastructure of muscles were observed by TEM. Result The results showed that Des, Vim and ultrastructure of SE had no significant changes compared with controls. But after SCDE, Des and Vim of smooth muscle decreased and ultrastructure had damaging changes. Conclusion Swimming exercises with different time spans will have different effects on medianfiber and ultrastructure of smooth muscle, and IF and ultrastructure of smooth muscle have different change on the different time spans of swimming exercises.
