不同电学参数电致孔时对栀子提取液黏膜给药的影响

摘要 目的:探索电致孔条件下不同电学参数(电压、波形、脉冲宽度、脉冲次数)对栀子提取液透黏膜给药的影响。方法:用中药电致孔给药仪产生多种电学参数条件,并以双室扩散池法、HPLC法检测栀子苷透过直肠黏膜的变化。结果:栀子苷的累积透过量与电参数密切相关,在单因素影响中,4个最佳电学参数分别为100V电压、方波波形、500Ns的脉冲宽度、360个脉冲次数,透过量有显著的增大。结论:优化的电致孔条件应用于中药有效成分透直肠黏膜给药有良好的前景。 OBJECTIVE To observe the effect of different electrical parameters (voltage, shap of wave, pulse width, and number of pulses) on rectum mucous membrane delivery of gardenia glucoside using eletroporation. METHODS The rectum mucous membrane penetration in Franz diffusion cell for isolated pig rectum mucous membrane of gardenia glueoside was studied by different electrical parameters of electroporation equipment for Chinese medicine, respectively. The content of gardenia glucoside permeated on rectum mucous membrane of pig was determined by HPLC. RESULTS The accumulative permeation of gardenia glucoside was correlated with electrical parameters, different electrical parameters showed the significant effect on gardenia glucoside' s rectum mucous membrane delivery. The optimum electrical parameters were voltage: 100 V, shap of wave: square wave,pulse width: 500 Ns, number of pulses: 360. The permeation increased significantly compared with the passive diffusion. CONCLUSION Optimum parameters of eletroporation have an favorable future for active constituents in Chinese materia medica to be rectum mucous membrane deliveried.
