摘要
目的:研究重组大肠杆菌ZJGSU01高效表达内切几丁质酶的最优化培养条件。方法:以内切几丁质酶活力为指标,对诱导剂的诱导时间、诱导剂的添加量、装液量和接种量进行优化。结果:接种量5%,装液量100mL/500mL,重组菌培养1h,添加诱导剂IPTG的浓度为1.0mmol/L,有利于内切几丁质酶的表达。结论:通过上述条件的优化,可明显提高内切几丁质酶的表达量。
In order to study the optimal culture conditions of the recombinant Escherichia coli ZJGSU01 for highly expressing endochitinase, the inducer adding time, inducer concentration, medium volume and inoculative amount were optimited, using endochitinase activity as index. The results indicated that inoculative amount of 5%, medium volume of 100 mL/500 mL, IPTG concentration of 1.0 mmol/L and inducer adding time of lh were beneficial to the endochitinase expression.
