摘要 目的制备5-氟尿嘧啶凝胶微球实现结肠定位治疗的目的。方法选择滴制法制备海藻酸钙凝胶微球,并选择丙烯酸树脂水分散体为衣膜材料,采用流化床技术包衣,以微球圆整度、包封率、释放度为质量指标,对球芯滴制工艺和处方进行了单因素考察,通过正交设计筛选出最优处方。结果凝胶微球体外4h药物累积释放率〈15%,12h〉85%。结论按最优化处方工艺制得的5-氟尿嘧啶微球符合结肠定位制剂要求,且性质稳定。 Objective To prepare 5-FU loaded gels microspheres for colon-specific. Methods The calcium alginatc gels microspheres were prepared by drop method. The orthogonal test was used to optimize the core formulation, and the coating process was performed on a mini-fluidized bed spray coater with Eudragit S100. Results The drug releasing percentage of the coated gel beads was less than 15% in 4 hours and reached 85% in 12 hour. Conclusion The optimized method can be used to prepare 5-FU colon-specific gel microsphere successfully.
