摘要
目的:观察丹参饮对心肌细胞凋亡与相关蛋白表达干预作用。方法:采用结扎冠状动脉左前降支的方法复制急性心肌缺血模型,15条犬随机分成对照组、心肌缺血组以及丹参饮组,每组5条。分别采用HE染色、DNA末端标记法(TUNEL)以及免疫组化法观察心肌组织病理变化、心肌细胞凋亡与相关蛋白表达的变化。结果:对照组无细胞变性与坏死,仅见极少量凋亡阳性细胞;心肌缺血组有明显心肌组织坏死,心肌细胞凋亡增多,Bcl-2的表达明显减少,Bax的表达明显增多,与对照组比较差异显著;丹参饮组心肌细胞坏死及凋亡均显著减轻,Bcl-2的表达明显增多,Bax的表达明显减少,与心肌缺血组比较差异显著。结论:丹参饮可影响Bcl-2和Bax的表达,有效地抑制心肌细胞坏死及凋亡,减轻心肌细胞损伤,对缺血心肌有保护作用。
Objective :To explore the intervention of Danshenyin decoction on myocardial cellular apoptosis and expressions of Bcl - 2, Bax in acute experimental ischemie myocardium. Methods : Fifteen dogs were divided into 3 groups randomly as control group ,isehemie group and Danshenyin group. The left anterior descending coronary artery was ligated to make a model of acute ischemic myocardium. The changes in myocardial pathology, cellular apoptosis and expressions of Bcl - 2, Bax were detected. Results : Cellular degeneration and necrosis were not fotmd in control group. Danshenyin decoction decreased myocardium cellular apoptasis and expression of Bax as well as increased expression of Bcl -2. Comparing with ischemie group ,it is significant( P 〈 0. 05). Conclusion: Danshenyin decoction can affect expressions of Bcl- 2, Bax and prevent myocardial cells from apoptosis.
